Influence of Rutin, Sinapic Acid, and Naringenin on Binding of Tyrosine Kinase Inhibitor Erlotinib to Bovine Serum Albumin Using Analytical Techniques Along with Computational Approach

Flavonoid-containing food supplements are widely used as antioxidants, and the continuous use of these with other drugs can lead to clinically significant interactions between drugs. The medications in systemic circulation mainly transported by serum albumin, a major transport protein. This study evaluated rutin (RUT), naringenin (NAR), sinapic acid (SIN) most abundant protein, bovine albumin (BSA), anticancer drug, tyrosine kinase inhibitor Erlotinib (ETB), using various analytical methods. Interaction multiple types ligands proteins competition themselves bound ETB’s displacement from BSA-binding site, leading elevated ETB concentrations circulation. These drug fractions adverse events lower tolerance, increased resistance therapeutic regimen ETB. experimental computational methods, including molecular-docking studies, were understand molecular interactions. results suggested that complexes formed utterly different binary ternary system. Furthermore, comparing systems amongst themselves, spectra differed each other. They thus inferred BSA-ETB presence RUT, NAR, SIN separately also different, highest value reduction binding energy followed then NAR. Thus, we conclude competitive flavonoids might influence pharmacokinetics cancer patients increasing tolerance or resistance.
